You only need one ISBN. The only time you would use another is for a different version like a Spanish or French translation version, or you did something like offer a special edition version with a different price or something like a Hardback offering. Even if you change the interior to add sample chapters or update the cover, you don't need a new ISBN.
Bowker will tell you to use a new one for everything, they are also the ones selling them for $125 a pop.
